The NHS Foundation funded a series of clinical trials to test the efficacy of sildenafil (Viagra®) in the treatment of erectile dysfunction. The trial was designed to test the drug sildenafil, a non-Sildenafil, which has been associated with an increased risk of sexual dysfunction.
The trial was a prospective, double-blind, placebo-controlled trial, in men who were prescribed sildenafil, a Sildenafil-only drug, to help men with erectile dysfunction, as a part of the treatment for a chronic condition called ‘erectile dysfunction’. The trial was run between 12 and 25 February 2024 at a high-risk population in Northern Ireland, and involved over 150,000 men and women. The men were assigned to one of four treatment groups (Group 1, 2, 3 and 4). To demonstrate the efficacy of sildenafil, the primary endpoint of the trial was measured in men who reported having erections of at least 20° or larger during the previous month. The trial had an average follow-up of 5.3 years.
For the primary study measure of the percentage of men in the treatment group who reported having erections of at least 20° or larger during the previous month, sildenafil was assessed at the end of the study period, in men who had at least one sexual activity at the time of randomisation.
For the secondary measure of the percentage of men in the treatment group who reported sexual satisfaction, sildenafil was assessed at the end of the study period, in men who had at least one sexual activity during the previous month, or had a PDE5 inhibitor, or a PDE5 inhibitor-only (Tadalafil®) group. All the secondary endpoints were analysed using independent-samplet-tests.
The secondary endpoints of the study were analysed using a multivariable logistic regression model to determine if the number of sexual activities or the percentage of the men in the treatment group who reported having erections of at least 20° or larger during the previous month, or with a PDE5 inhibitor, or a PDE5 inhibitor-only (Tadalafil®) group differed from the total number of sexual activities or the percentage of the men in the treatment group who reported sexual satisfaction or the percentage of the men in the treatment group who reported sexual satisfaction during the previous month.
A sensitivity analysis was performed to determine whether the number of sexual activity during the previous month of the treatment group also differed from the total number of sexual activity or the percentage of the men in the treatment group who reported sexual satisfaction.
The drug, which was sold on the internet in the UK, is said to be the first medication approved by the Food and Drug Administration in the country.
